The web site won't let you put more than 2 people per room, but it does not mean you won't fit. We got a room with 2 double beds, and it fit three adults. It is a tight squeeze with two two adults in one bed. You should email the hotel to see if they have cribs. Towards the window there is a chair and a small table, that you can move to make room for the crib. The room is small by western standards, but big for Japanese standards. We stayed there because it had the biggest rooms I could find. It is next to a Shinkansen station and also a metro station. I would not recommend trying to use he subway with 2 kids. The train/metro stations are quite crowded and chaotic. Taxis will be easier but more expensive. Also consider how you are going to get from the airport to the hotel. It is bad enough dragging your luggage through the train station as adults, but add in a toddler and a baby is something else. Can can save considerable trouble by shipping your luggage from the airport to the hotel, it will arrive he next day, and is not expensive. Just bring enough luggage with to the hotel for the first night. Airports will have luggage delivery services.
A mentioned by another member, this hotel and train station is not really in the middle of town. The Marriott is at the Shin-Osaka station, the middle of town is Osaka station, so it is confusing. Osaka station is only a fee metro stops from Shin-Osaka. I don't remember every seeing a baby stroller in the subway station or even out in the streets. If you have never been in Japan before, you will be shocked at how crowded it is. The train/metro stations are also not easy to navigate with a stroller. You really have to hunt for a elevator, and not all stations have elevators, and everybody is walking at a fast pace.
