Edited: 11 years agoThere are direct trains from Florence to Perugia every 2 hours, taking just over 2 hours for the journey and costing €12.35. There is no point trying to buy tickets in advance. These are Regionale trains with no seat reservations (so tickets cannot sell out) and no discounts.
But why not pick up the car in Florence and drive to Perugia? It is under 100 miles and for this journey a car would be faster than a plane. If you are worried about traffic fines in Florence, take a taxi to the airport (€20) and pick up the car there.
There will be a wider range of cars in Florence, and this could be important if you want an automatic.
Depending where in Umbria you plan to go, there are also very good public transportation options out of Perugia to many neighboring towns and villages (bus and train, which are even more attractive if you're travelling solo):
We based ourselves in Perugia to tour Umbria a couple of years ago. It is a great town to stay in. We flew into Rome so picked up our car at the airport to drive to Perugia. It is not a difficult drive.
As sydneynick suggests, you may have a better choice of cars in Florence. It is not always easy to get an automatic in Italy even if you have pre-booked!
We rented from Avis and they have an office right at the railway station in Perugia. We returned our car there before taking the train to Florence.
